These are large, fragile protein molecules that the digestive system would ordinarily destroy before they could reach the bloodstream. The oral semaglutide solution involved co-formulating the peptide with SNAC (sodium N-[8-(2-hydroxybenzoyl)aminocaprylate), an absorption enhancer that protects semaglutide from enzymatic breakdown in the stomach and facilitates transcellular absorption through the stomach wall. When patients stayed on treatment for the full duration, that number climbed to 16.6%, essentially matching the weight loss performance of injectable WEGOVY 2.4 mg.<\/p>\n\n\n\n<p>The WEGOVY pill isn&#8217;t just indicated for weight loss. FOUNDAYO, by contrast, can be taken at any time of day, with or without food, with or without water. No fasting. No timing rules. Just take it. This pharmacological advantage could prove decisive in the real-world adherence battle.<\/p>\n\n\n\n<p>Clinically, the <strong>ATTAIN Phase 3 program<\/strong> showed that orforglipron at its highest dose delivered a mean weight loss of 12.4% over 72 weeks, slightly less than the WEGOVY pill&#8217;s 13.6% at 64 weeks, but with a broader, more diverse patient population of over 3,000 participants. More than 54.6% of participants achieved \u226510% weight loss, alongside meaningful improvements in cardiometabolic markers, including blood pressure, waist circumference, and lipid levels.<\/p>\n\n\n\n<p>Eli Lilly priced FOUNDAYO comparably to the WEGOVY pill, with self-pay access starting at $149\/month for the lowest dose, eligible commercial insurance patients paying as low as <strong>$25\/month<\/strong> with savings cards, and Medicare Part D patients expected to access it for <strong>$50\/month<\/strong> beginning <strong>July 1, 2026<\/strong>.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"h-the-oral-glp-1-pipeline-more-challengers-are-coming\"><span class=\"ez-toc-section\" id=\"The_Oral_GLP-1_Pipeline_More_Challengers_Are_Coming\"><\/span><strong>The Oral GLP-1 Pipeline: More Challengers Are Coming<\/strong><span class=\"ez-toc-section-end\"><\/span><\/h2>\n\n\n\n<p>The <a href=\"https:\/\/www.delveinsight.com\/blog\/obesity-drugs-launch\">obesity drug pipeline<\/a> is one of the most dynamic and rapidly evolving areas in modern medicine, with <strong>more than 100 candidates<\/strong> in development targeting diverse physiological pathways. While the currently approved GLP-1 therapies have transformed obesity management, they represent only the first wave of innovation. A robust pipeline of oral GLP-1 agents and next-generation therapies with complementary mechanisms is advancing swiftly, intensifying competition and expanding future treatment possibilities.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-pfizer-s-pf-07976016\"><span class=\"ez-toc-section\" id=\"Pfizers_PF-07976016\"><\/span><strong>Pfizer&#8217;s PF-07976016<\/strong><span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p>Pfizer was once the most aggressive challenger in the oral GLP-1 race. Its small-molecule candidate danuglipron showed promising efficacy, with placebo-adjusted weight reductions of 8\u201313% in Phase 2b trials. But the path to approval was rocky. High gastrointestinal discontinuation rates exceeding 50% across all doses, combined with liver safety signals in early studies, ultimately led Pfizer to discontinue danuglipron&#8217;s development in April 2025. The company, however, has not abandoned the space. It is continuing the development of an oral GIPR antagonist candidate (PF-07976016) in Phase 2, and maintains an earlier-stage obesity pipeline.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\" id=\"h-regor-therapeutics-rgt-075\"><span class=\"ez-toc-section\" id=\"Regor_Therapeutics_RGT-075\"><\/span><strong>Regor Therapeutics\u2019 RGT-075<\/strong><span class=\"ez-toc-section-end\"><\/span><\/h3>\n\n\n\n<p>RGT-075 is a once-daily oral small-molecule GLP-1 receptor full agonist being developed by Regor for the treatment of metabolic disorders such as <a href=\"https:\/\/www.delveinsight.com\/report-store\/type-2-diabetes-market\">type 2 diabetes mellitus<\/a>, obesity, and overweight conditions linked with weight-related comorbidities. The candidate has previously advanced through Phase IIa clinical evaluation in obese patients, as well as Phase I single ascending dose (SAD) studies in healthy volunteers and multiple ascending dose (MAD) studies in diabetic patients.
