LUMAKRAS Market Summary

Key Factors Driving LUMAKRAS Growth

1. Major expansion into KRAS G12C-mutated colorectal cancer: The biggest recent growth driver is the FDA approval in January 2025 of LUMAKRAS in combination with panitumumab for adults with KRAS G12C-mutated mCRC after prior fluoropyrimidine-, oxaliplatin-, and irinotecan-based chemotherapy. The approval was based on CodeBreaK 300, where sotorasib 960 mg + panitumumab produced median PFS of 5.6 months versus 2.2 months with investigator's-choice therapy (HR 0.49) and an ORR of 26.4% versus 0%. This creates a second commercially important tumor type for LUMAKRAS beyond NSCLC.

2. Large unmet need in refractory KRAS G12C-positive mCRC: KRAS G12C mutations occur in approximately 3% of colorectal cancers, and metastatic CRC has a poor prognosis, with a reported 5-year relative survival of only 15.7%. The FDA therefore recognizes KRAS G12C-mutated refractory CRC as a serious condition with limited treatment options. LUMAKRAS' ability to provide a biomarker-directed option after standard chemotherapy supports continued uptake in this population.

3. Combination with panitumumab improves the effectiveness of KRAS inhibition: LUMAKRAS' activity in CRC is strengthened by combining it with EGFR blockade. In CodeBreaK 300, the 960-mg combination more than doubled median PFS compared with investigator's choice, while the response rate was 26% versus 0%. Updated overall-survival analysis showed an OS HR of 0.70, although the study was not powered to demonstrate a statistically significant OS difference. These results support the combination as an important post-chemotherapy targeted-treatment option.

4. Established efficacy and durable responses in KRAS G12C-positive NSCLC: LUMAKRAS remains an established treatment for previously treated KRAS G12C-mutated advanced NSCLC. In the CodeBreaK 100 Phase II study, the ORR was 37.1%, median duration of response was 11.1 months, disease-control rate was 80.6%, and median overall survival was 12.5 months. A longer follow-up analysis reported an ORR of 40.7%, median PFS of 6.3 months, median OS of 12.5 months, and 32.5% of patients alive at two years.

5. Growing international contribution is supporting revenue: Amgen's actual sales data show particularly strong growth outside the U.S. In 2025, LUMAKRAS/LUMYKRAS generated $363 million globally, compared with $350 million in 2024, representing 4% growth. U.S. sales were essentially flat at $211 million versus $214 million in 2024, while ROW sales increased 12% to $152 million. Amgen attributed the 2025 full-year increase primarily to 13% volume growth, partially offset by lower net selling price. This suggests that geographic expansion and patient-volume growth remain important commercial drivers.

LUMAKRAS Recent Developments

In January 2025, Amgen announced that the US Food and Drug Administration (FDA) had approved LUMAKRAS (sotorasib) in combination with Vectibix (panitumumab) for the treatment of adult patients with KRAS G12C-mutated metastatic colorectal cancer (mCRC), as determined by an FDA-approved test, who have received prior fluoropyrimidine-, oxaliplatin- and irinotecan-based chemotherapy. Approval is based on the pivotal Phase III CodeBreaK 300 study, which demonstrated that LUMAKRAS plus Vectibix is the first and only targeted treatment combination for chemorefractory KRAS G12C-mutated mCRC to show superior progression-free survival (PFS) compared to the investigated standard-of-care (SOC).

LUMAKRAS Drug Summary

LUMAKRAS (Sotorasib) is an orally administered, small-molecule inhibitor of the KRAS G12C protein developed by Amgen for the treatment of cancers harboring the KRAS G12C mutation. By selectively and irreversibly binding to the cysteine residue created by the G12C mutation, sotorasib locks KRAS in an inactive state and suppresses downstream signaling pathways involved in tumor-cell proliferation and survival. LUMAKRAS is approved as a single agent for adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have received at least one prior systemic therapy, and in combination with panitumumab for adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) previously treated with fluoropyrimidine-, oxaliplatin-, and irinotecan-based chemotherapy. The recommended dose is 960 mg orally once daily, with treatment continued until disease progression or unacceptable toxicity. Common adverse reactions include diarrhea, musculoskeletal pain, nausea, fatigue, hepatotoxicity, and cough in NSCLC, while combination treatment in colorectal cancer commonly causes rash, dry skin, diarrhea, stomatitis, fatigue, and musculoskeletal pain; important serious risks include hepatotoxicity and interstitial lung disease/pneumonitis.
