In disease states, excessive TGF-β signaling promotes abnormal collagen deposition, fibroblast activation, tissue scarring, tumor progression, and immune evasion.

TGF-β inhibitors interfere with this cascade through multiple mechanisms, including neutralizing TGF-β ligands with monoclonal antibodies, preventing ligand–receptor interactions, inhibiting TGF-β receptor kinase activity using small-molecule inhibitors, or blocking downstream SMAD signaling. Some therapies also employ antisense oligonucleotides or ligand traps to reduce TGF-β expression or bioavailability.
